At the beginning of the movie, it is implied that the Relic containing the Blood of Jesus Christ is in the chambers beyond the corridor full of crosses and the door that says 'God Ends Here', and that the two sisters are trying to retrieve it to destroy Valak. However, in the climax, it is revealed that the chambers are where the portal to Hell is presumably, so it makes no sense to open the door.
