Khan's deceased wife whom Khan mentions is Lt. Marla McGivers. When Khan was revived by the Enterprise crew, Lt. McGivers was manipulated by Khan to help him hijack the Enterprise. After Khan's murderous plans were foiled, Lt. McGivers was court martialed for her actions and Lt. McGivers decided to go with Khan and his followers to Ceti Alpha V, which Khan accepted.
