The number 42 is referenced multiple times throughout the film, which takes place in Brooklyn, NY. New York was formerly home to Brooklyn Dodgers (now the Los Angeles Dodgers) and legendary baseball player Jackie Robinson. It's also directly lifted from Miles's origin story, where both the lottery ball that earns him a scholarship to Brooklyn Visions and the spider that eventually bites him and gives him his powers are both conspicuously marked "42". "42" is also widely known as the answer to the question of "Life, the Universe & Everything" from the Douglas Adams "Hitchhiker's Guide to the Galaxy" series and is frequently referred to in genre fiction and geek culture.
