It's revealed in Detective Monk: Mr. Monk and the Three Pies (2004) (#2.11) that Adrian Monk has a brother named Ambrose. It's revealed in Detective Monk: Mr. Monk's Other Brother (2009) (#7.10) that he has a paternal half-brother.
