After Neville faints in the greenhouse, Seamus Finnegan and Professor Sprout talk to each other without removing their earmuffs. If they can hear each other, they should have been able to hear the mandrakes screaming, and should have had some kind of a reaction to the sound. As explained elsewhere, the earmuffs may be enchanted to only block out the sound of the mandrakes and not other sounds.
