Frank Bullitt, tenente della squadra omicidi di San Francisco, viene incaricato da Walter Chalmers, un politicante, di sorvegliare in una stanza d'albergo un certo Johnny Ross, un elemento dell'organizzazione mafiosa, che, fuggito con alcuni milioni di dollari della stessa, è pronto a testimoniare contro i suoi capi. Nonostante tutte le precauzioni prese, durante una notte, per una leggerezza di Ross, che toglie la catena alla porta, due sicari entrano nella stanza e feriscono gravemente l'agente di guardia e il testimone. Mentre questi viene portato all'ospedale, Chalmers, vedendo sfuggire l'occasione per raggiungere i suoi scopi, esige, ma inutilmente, da Bullitt una dichiarazione di colpa.
