New York, 1933. Durante la Grande Depressione la fame di fama conduce una troupe cinematografica su un'isola misteriosa per girare un film spettacolare e avventuroso. A bordo della Venture salpano Carl Denham, regista ambizioso, Ann Darrow, un'attrice esordiente e Jack Driscoll, un drammaturgo intellettuale prestato al cinema. Durante una tempesta la nave si incaglia sugli scogli dell'Isola del Teschio, una terra fuori da ogni mappa e governata da un colossale scimmione, Kong, a cui viene offerta in sacrificio la bella Ann. Gli uomini della Venture, nel tentativo di salvare la fanciulla, cattureranno Kong e lo "trasferiranno" a Manhattan per esibirlo come fenomeno da fiera. Ma Re Kong spezzerà le catene e scalerà l'Empire State Building ghermendo di nuovo la bella dai capelli d'oro.
