La piccola principessa Dala riceve dal padre un enorme diamante, la Pantera Rosa, al cui interno sembra incisa una pantera saltellante. I ribelli prendono il controllo del paese e Dala fugge col diamante. Qualche tempo dopo, ormai bellissima donna, a Cortina d'Ampezzo, incontra sir Charles Lytton, ladro gentiluomo insieme col nipote americano George, che ambisce come lo zio alla preziosa pietra. A seguirli l'Ispettore Clouseau e sua moglie Simone, segreta amante e complice di Lytton. Simone tenta così, nonostante la presenza del marito, di evitare George, innamorato di lei, e di aiutare Charles. In una festa in costume, questa volta a Roma nella villa di Dala, i due ladri tentano di rubare il diamante, che è sparito, venendo colti in flagrante da Clouseau, che dà inizio ad una frenetica corsa con le auto per la città. Così, i due ladri vengono catturati ma alla fine, a causa di un intrigo ordito da Simone con la complicità di Dala, Clouseau diventa da accusatore ad accusato.
