Quando l'Enterprise fa ritorno sulla Terra in seguito ad un controverso incidente galattico, l'indomito Capitano Kirk è impaziente di tornare fra le stelle per una nuova e più lunga missione di pace ed esplorazione. Le cose però non vanno affatto bene sul Pianeta Blu. Un devastante attacco terrorista ha rivelato una realtà allarmante e la Flotta Stellare ha subìto un attacco dall'interno dalle disastrose conseguenze globali. Il Capitano Kirk guiderà l'Enterprise in una missione incredibile che spazia dal pianeta Klingon alla Baia di San Francisco. Il nemico che si annida a bordo dell'Enterprise ha un unico scopo: la distruzione totale. Kirk guiderà i suoi compagni in un ingannevole regno di specchi dove non si erano mai inoltrati prima d'ora, penetrando il sottile confine fra amici e nemici, vendetta e giustizia, alle prese con una guerra totale e la potenzialità di un futuro unito.
